Nucleic acid delivery to specific tissues and cells is very challenging. To address the delivery barriers, a variety of nanoparticle formulations, including lipid nanoparticles, have been tested. In this presentation, we will focus with case studies on endogenous mechanisms that can enhance nucleic acid delivery to their target – especially extrahepatic delivery.

Three strategies - role of protein corona, use of cells as Trojan Horse, and extracellular vesicles – will be discussed.
